Opal Polaris 780 Reagent Pack

Opal™ is a practical workflow for simultaneous detection of up to 8 tissue biomarkers plus nuclear counterstain, within a single image. The method is similar to standard immunohistochemistry and is accessible to many laboratories where standard IHC method development is performed.

Detail Information

The Opal Polaris 780 Reagent Pack is a two-part reaction and part of the Phenoptics 2.0 7-color whole slide workflow. The Reagent Pack contains:

  • Opal TSA-DIG, 1 vial
  • Opal Polaris 780, 1 vial
  • Dimethyl sulfoxide (DMSO), 1 x 100 uL

Users should source primary antibodies and other workflow reagents separately. Pack contains enough reagent to stain 50-slides. Generally, 100-300 µL of Opal Working Solution is required per slide.
Protocol for Opal TSA-DIG reconstitution and dilution:

  1. Dissolve the Opal TSA-DIG Reagent in 75 uL of DMSO.
  2. Before each procedure, dilute Opal TSA-DIG in 1X Plus Amplification Diluent for manual staining or 1X Plus Automation Amplification Diluent for automation to make Opal TSA-DIG Working Solution. The recommended dilution ratio is 1:100.

Protocol for Opal Polaris 780 reconstitution and dilution:

  1. Dissolve the Opal Polaris 780 Reagent in 300 uL of ddH2O.
  2. Before each procedure, dilute Opal Polaris 780 in Blocking/Ab Diluent to make Opal Polaris 780 Working Solution. The recommended dilution ratio is 1:25.

The Opal Reagent Packs can be utilized for both mono- and multiplex tissue staining and have been configured and validated for imaging on the Vectra® Polaris™ quantitative pathology imaging system.

Note: If you have never used Opal Multiplex IHC Detection Reagents before, we strongly suggest purchasing the full Opal kit, which includes a set of quality controlled reagents that have been optimized to work together.
Opal Multiplex IHC Kits make multiplex methods accessible to anyone who works with standard immunohistochemistry. The Opal method allows use of any number of unlabeled primary antibodies from the same species in multiplexed tissue assays, with no fear of cross-reactivity. Antibodies for simultaneous IHC may be selected based on performance, rather than species. The method works with formalin fixed paraffin-embedded (FFPE) tissue and is compatible with the standard research IHC workflow in your lab.
