PhenoCycler Assays – Reagents and Antibodies

The PhenoCycler® System (formerly CODEX®) offers a complete solution that includes antibodies and reagents optimized and validated for the PhenoCycler System. We now provide an expanded menu of gold standard verified antibodies containing essential and critical immune markers ideal for rare cell discovery, spatial neighborhood analysis, and more.

Our inventoried antibodies are validated for different tissue types including but not limited to; mouse fresh frozen, human fresh frozen and human formalin-fixed paraffin-embedded (FFPE) tissue. Users have the flexibility to create panels comprised of commercially available, ready-to-use Akoya-validated antibodies. Custom conjugation kits are also available for labeling antibodies with PhenoCycler barcodes. These ready-to-use reagents coupled with the fastest single-cell spatial biology system enable rapid and deep spatial phenotyping across whole tissue at scale.

Kits & Reagents

The PhenoCycler System requires specially formulated reagents and buffers for tissue staining with PhenoCycler antibodies and running iterative imaging cycles. For each imaging cycle, three PhenoCycler reporters, each with a spectrally distinct dye, are applied to the stained tissue to visualize the corresponding PhenoCycler antibody. The PhenoCycler instrument applies gentle wash steps to remove the PhenoCycler reporters without any chemical modifications, followed by applying the next set of Reporters to visualize the next set of PhenoCycler antibodies.

PhenoCycler Antibodies

PhenoCycler antibodies are provided individually and can be tailored to design panels for the simultaneous analysis of100+ biomarkers. Akoya provides fully validated PhenoCycler antibodies and a list of supplemental antibodies that have undergone varying levels of testing.

PhenoCycler Inventoried Antibodies

PhenoCycler inventoried antibodies are supplied by Akoya and have passed the PhenoCycler antibody validation process and manufacturing quality control. Our inventoried antibodies are also fully supported by our field support team.

PhenoCycler antibodies have been tested in multiple types of human FFPE tissue. These PhenoCycler antibodies are conjugated to a distinct PhenoCycler Barcode (BX000). Each kit contains one tube of PhenoCycler antibody and one tube of the corresponding dye-labeled Reporter (RX000), sufficient volume for 25 PhenoCycler runs (or tests) at the standard staining conditions. Recommended dyes for FFPE tissue are Atto 550, Cy5 or Alexa Fluor™ 647, and Alexa Fluor™ 750.
